چکیده

Acknowledgment The current work was carried out at the Department of Biological Chemistry of the University of Konstanz during the period of 1999-2002. Working there was at the same time instructional and fun, both of which being constantly upheld by my supervisor Prof. Dr. Volker Ullrich. Besides instructions, I like to thank him for his excellent support and working facilities he provided. I am grateful to all of my colleagues and co-workers for their friendship, collaboration and support especially to Dr. also my diploma candidates for their very dedicated cooperation and the numerous I had the privilege to become a member of the " Graduiertenkolleg Biochemische Pharmakologie " and I profited very much from the collaborations and support among the members, its seminars, its courses and the attendance of national and international congresses. I wish to thank the organisers Prof. Prof. Dr Ullrich for their professional running the " Kolleg ". Last and final thanks go to my beloved family.

Aldosterone Induces Oxidative Stress Via NADPH Oxidase and Downregulates the Endothelial NO Synthesase in Human Endothelial Cells

Aldosterone is traditionally viewed as a hormone regulating electrolyte and blood pressure homeostasis. Recent studies suggest that Aldo can cause microvascular damage, oxidative stress and endothelial dysfunction. However, its exact cellular mechanisms remain obscure. This study was undertaken to examine the effect of Aldo on superoxide production in human umbilical artery endothelial cel...

Ceramide-induced impairment of endothelial function is prevented by CuZn superoxide dismutase overexpression.

OBJECTIVE Ceramide is an important intracellular second messenger that may also increase superoxide. The goal of this study was to determine whether overexpression of CuZn superoxide dismutase (SOD) protects against ceramide-induced increases in vascular superoxide and endothelial dysfunction. METHODS AND RESULTS Carotid arteries from CuZnSOD-transgenic (CuZnSOD-Tg) and nontransgenic litterma...

Low NO bioavailability in CCl4 cirrhotic rat livers might result from low NO synthesis combined with decreased superoxide dismutase activity allowing superoxide-mediated NO breakdown: A comparison of two portal hypertensive rat models with healthy controls

BACKGROUND: In cirrhotic livers, the balance of vasoactive substances is in favour of vasoconstrictors with relatively insufficient nitric oxide. Endothelial dysfunction has been documented in cirrhotic rat livers leading to a lower activity of endothelial nitric oxide synthase but this might not be sufficient to explain the low nitric oxide presence. We compared the amount of all nitric oxide ...

Comparing the effect of 6 weeks of continuous and interval aerobic training on vascular endothelial growth factor and superoxide dismutase enzyme in hippocampus of male rats of Parkinson's model

Introdution: Based on researches, increased levels of vascular endothelial growth factor and antioxidant parameters in areas of the brain that are more affected by Parkinson's disease have a potential therapeutic effect. Therefore, the purpose of this study was to compare the effect of 6 weeks continuous and interval aerobic exercises on vascular endothelial growth factor and superoxide dismuta...

Selective and sensitive electrochemical biosensing of superoxide anion production by biological systems: a short overview of recent trends

Biologically, superoxide anion (O2 ) has been proposed to be directly or indirectly involved as a signaling messenger in vascular dysfunction [1,2]. An overproduction has been observed during ischemia and reperfusion and in pathologies such as hypercholesterolemia, atherosclerosis and diabetes. The superoxide anion radical belongs to reactive oxygen species and may damage organisms if it exceed...

Nephrotoxicity of Isosorbide Dinitrate and Cholestasis in Rat: The Possible Role of Nitric Oxide

Background: Nitric oxide (NO), a major chemical form of endothelium-derived relaxing factor and an important regulator of vascular tone, is released by endothelial cells. The role of NO is not restricted to the vascular system, and it participates in the regulation of renal hemodynamics and renal excretory function. There are increasing evidences indicating that the elevated levels of NO play a...
